Four challenges we have to crack before immune therapy can revolutionize how we fight cancer

Sri Krishna, Arizona State University Most of us know about the conventional treatment of cancer: surgery to remove tumors, chemotherapy and radiation. But within the last five years, a new class of drugs that use our immune systems to fight cancer are gaining traction in cancer treatment. This is called “immunotherapy.” Instead of killing cancer cells with radiation or chemotherapy, immunotherapy mobilizes the immune system to fight against cancer much like it does against bacteria and viruses. For a training immunologist like myself, immune therapies have opened new doors to understanding and treating cancers. In the future, immunotherapy could mean a personalized treatment, entirely tailored to an individual. As exciting as that sounds, we still have plenty of work to do, as there remains a lot we don’t know about the immune system. Here are some of the challenges we need to overcome to create these personalized treatments. Challenge #1: Tumors don’t want the immune system to know they are tumors In the late 1980s, researchers started to wonder if the immune system could fight tumors the same way it fought foreign invaders. Studies in mice and in humans showed that when immune cells were extracted from a body, reprogrammed and transferred back into the same body, they caused tumors to temporarily shrink, giving first indications of immunotherapy at work. In turns out that tumors are pretty vicious in avoiding the immune system. In the 1990s, researchers discovered that tumors can actively put “brakes” on the body’s immune system to evade detection by T-cells, a special kind of immune cell. T-cells search for signs of infection within cells by looking for specific protein codes on the cell surface. When T-cells decide that protein codes displayed on a cell don’t look like they belong to normal cells, they attack these abnormal cells and kill them (as shown in this fantastic video). Tumors can block T-cells from recognizing these protein codes. By doing this, tumors essentially go into stealth mode, flying under the radar of immune detection. We’ve figured this challenge out – sort of. Extensive research on the T-cell brakes eventually led to drugs that block the effects of these brakes – allowing the immune system to detect and attack tumors that would otherwise be invisible to it – and present-day cancer immunotherapy was born. Curcumin improves memory and mood, new UCLA study says

Lovers of Indian food, give yourselves a second helping: Daily consumption of a certain form of curcumin — the substance that gives Indian curry its bright color — improved memory and mood in people with mild, age-related memory loss, according to the results of a study conducted by UCLA researchers. The research, published online Jan. 19 in the American Journal of Geriatric Psychiatry, examined the effects of an easily absorbed curcumin supplement on memory performance in people without dementia, as well as curcumin’s potential impact on the microscopic plaques and tangles in the brains of people with Alzheimer’s disease. Found in turmeric, curcumin has previously been shown to have anti-inflammatory and antioxidant properties in lab studies. It also has been suggested as a possible reason that senior citizens in India, where curcumin is a dietary staple, have a lower prevalence of Alzheimer’s disease and better cognitive performance. “Exactly how curcumin exerts its effects is not certain, but it may be due to its ability to reduce brain inflammation, which has been linked to both Alzheimer’s disease and major depression,” said Dr. Gary Small, director of geriatric psychiatry at UCLA’s Longevity Center and of the geriatric psychiatry division at the Semel Institute for Neuroscience and Human Behavior at UCLA, and the study’s first author. The double-blind, placebo-controlled study involved 40 adults between the ages of 50 and 90 years who had mild memory complaints. Participants were randomly assigned to receive either a placebo or 90 milligrams of curcumin twice daily for 18 months. All 40 subjects received standardized cognitive assessments at the start of the study and at six-month intervals, and monitoring of curcumin levels in their blood at the start of the study and after 18 months. Thirty of the volunteers underwent positron emission tomography, or PET scans, to determine the levels of amyloid and tau in their brains at the start of the study and after 18 months. The people who took curcumin experienced significant improvements in their memory and attention abilities, while the subjects who received placebo did not, Small said. In memory tests, the people taking curcumin improved by 28 percent over the 18 months. Those taking curcumin also had mild improvements in mood, and their brain PET scans showed significantly less amyloid and tau signals in the amygdala and hypothalamus than those who took placebos. What is kombucha and how do the health claims stack up?

Kombucha is made from either a green or black tea base, with some interesting additions. Matt Hoffman Evangeline Mantzioris, University of South Australia and Permal Deo, University of South Australia The drink kombucha was previously only popular in hipster cafes, but is now vying for space on the supermarket shelves. Many claims are made about the health benefits of drinking kombucha, but what does the science say? For those of you who haven’t tried it, kombucha is a quirky-tasting drink. Depending on what’s added to it, it’s best described as a sour apple cider, perhaps with vinegary notes. Kombucha is an ancient beverage that was originally consumed in China more than 2,200 years ago for its detoxifying and energising properties. As trade routes expanded, kombucha found its way to Russia and then into other eastern European areas. During the second world war, kombucha was introduced into Germany, and in the 1950s it became popular in France and North Africa. By the 1960s, Swiss scientists claimed that kombucha was beneficial for the gut in a similar way to yoghurt. How it’s made Kombucha is made from either a green or black tea base. Added to that is white sugar, which has been fermented with a type of “tea fungus” called a symbiotic culture of acetic acid (vinegar) bacteria and yeast, or SCOBY, for one to two weeks. Detailed scientific recipes are available for how to prepare kombucha. The taste of the kombucha changes during fermentation from a pleasantly fruity sour-like sparkling flavour, to a mild vinegary taste after a long incubation period. The fermentation process is important, as the SCOBY changes the polyphenols – compounds normally found in tea, fruits and vegetables – into other organic compounds. This increases the acidity, which prevents other micro-organisms growing. It is these new organic compounds that are claimed to provide health benefits beyond those already found in green or black tea. The fermentation process The fermentation process extends the shelf-life of kombucha, as it does with other historically popular foods and drinks such as cheese, yoghurt, sauerkraut, kimchi, wine and beer. Beer and wine become alcoholic during the fermentation process, whereas kombucha generally contains less than 0.5% alcohol. Food Standards Australian and New Zealand classifies foods and drinks below 0.5% as non-alcoholic. Ethanol, the alcohol you can drink, is produced when the yeasts and bacteria in the SCOBY interact via a process called glycolysis.

Jyoti Singh was a 23-year-old medical student when she was gang raped on a bus in India in 2012 before dying from her injuries, igniting an international firestorm about crimes against women. Four attackers received the death penalty and the Indian government created fast track courts to hear rape cases. Like many others, India native Poorvi Mathur was left distraught by the story. “It shook a lot of us in India,” said Ms. Mathur. “I wanted to do something about it.” What the former Monroeville mother of two young daughters eventually did was help form a biotech startup company that is designing a device intended to make women safer. Ms. Mathur has since moved back to India with her family, while the family weighs plans for a return to the U.S., but the company’s three co-founders all have Pittsburgh-area ties, even though they don’t yet have a corporate office and use borrowed lab space in Oakland to test their hardware. The early stage company — Hera Global Tech Inc. — takes its name from the wife of Zeus, queen of the ancient Greek gods. It was formed in 2017 and is funded, for now, by family and friends. The total investment so far was not disclosed. But Hera Global has big funding plans for its formal launch: the company is a finalist in the Anu and Naveen Jain Women’s Safety Xprize, which is sponsored by the Culver City, Calif.-based Xprize Foundation. The winner, which will be announced in June, gets $1 million. In the meantime, the company is scrambling to develop a wearable, waterproof device — about the size of a matchbook — that can detect danger and alert emergency contacts. The idea is to measure heart rate and other metrics that indicate apprehension, signaling danger. The device could be worn by anyone, including firefighters and first responders, but women will be the company’s first target market. Under one of the contest rules, the device can cost no more than $40, which the co-founders believe is achievable. “I’m not nervous about it,” said Elizabeth LaRue, 46, a Hera Global co-founder and West Virginia native who received a doctorate from the University of Pittsburgh in library science and has years of experience teaching health informatics. “We’re making good steady progress.” Ms. Mathur, 37, a graduate of the Savannah College of Art and Design, said the device will have global positioning system

But while breast cancer survivors are grateful their bodies show no trace of the disease, they still face anxiety. Breast cancer can and does return, sometimes with a vengeance, even after being in remission for several years. By studying the “cannabilistic” tendency of cancer cells, my research team has made some progress in finding out why. The chances of recurrence and disease outcome vary with cancer subtype. About one-third of patients diagnosed with triple negative breast cancer, the most aggressive subtype, may experience a recurrence in another part of the body. This is called distant recurrence. It has been difficult, if not impossible, to predict if and when the same cancer will recur – and to stop it. Recurrent disease may arise from just a single cancer cell that survived the initial treatment and became dormant. The dormancy allowed it to hide somewhere in the body, not growing or causing harm for an unpredictable amount of time. Determining what puts these dormant cells to “sleep” and what provokes them to “wake up” and begin multiplying uncontrollably could lead to important new treatments to prevent a demoralizing secondary cancer diagnosis. Recently, my research team and I uncovered several clues that might explain what triggers these breast cancer cells to go dormant and then “reawaken.” We showed that cell cannibalism is linked to dormancy. How do bone stem cells affect breast cancer? Breast cancer can recur in the breast or in other organs, such as the lungs and bone. Where breast cancer decides to grow depends largely on the microenvironment. This refers to the cells that surround it, including immune cells, cells comprising blood vessels, fibroblasts and the select proteins they produce, among other factors. Over a century ago, a surgeon named Stephen Paget famously compared the organ-specific prevalence of cancer metastasis to seeds and soil. Because breast cancer often relapses in bones, in this metaphor, which still holds forth today, the bone marrow provides a favorable microenvironment (the “soil”) for dormant breast cancer cells (the “seeds”) to thrive. Crying elephants and giggling rats – animals have feelings, too

Indeed, a cow was just a walking burger, steak of Sunday roast, keeping itself fresh and tasty ready for when we were hungry. Luckily, for their sake, things have progressed significantly from then and now we recognise that animals (including our “superior” human selves in that category) can experience emotions from more simple ones such as happiness and sadness to more complex ones such as empathy, jealousy and grief. Animal sentience is defined as the ability to feel, perceive and experience subjectively. In other words, it’s about emotions and feelings and in some respects, having an awareness that “you are you”. In fact, the scientific evidence for animals being sentient is vast – so clear that three scientists read 2,500 papers studying sentience in non-human animals and concluded confidently that sentience does indeed exist. If you saw Blue Planet II recently, for example, you’ll have seen the footage of a pilot whale carrying around her dead calf. For most humans, this clearly demonstrates a form of grieving, particularly given the behaviour changes in the wider family pod. The evidence for sentience Studies have shown that sheep are able to recognise the faces of their sheep friends even after being separated for two years. Elephants from strong family groups with immense memories and they cry when they are hurt (both physically and emotionally). Capuchin monkeys know when they are receiving unequal pay (grapes vs cucumber) and Macaques develop individual cultures, particularly when it comes to how one should wash a potato. Chimpanzees like to keep the peace by redistributing bananas if someone complains that their share is unfair and even rats have been shown to demonstrate empathy by giving up their favourite snack to save a drowning friend. They also giggle when being tickled. Fish use tools and octopus weigh up whether the effort needed to gain a food reward is worth it depending on the type of food. There is also plenty of evidence on how animals have individual personalities and indeed how some are a glass half full type while others are more glass half empty. But it isn’t just from watching their behaviour that we can say animals are sentient. When we examine the brains of species (and indeed individuals), we can draw parallels

The three scientist’s lives were crowded with unmatched triumphs, reputation, and excellent human soul. We are memorising about India’s most exceptional scientists Dr. Lalji Singh (July.05, 1947 – Dec.10, 2017), Dr. K. Vijaya Raghavan (Oct.01, 1943 – Oct.12, 2017) and Dr. P. M. Bhargava (Feb. 22, 1928 – Aug. 01, 2017), who are no more with us today. The memorial gathering was seeking to portray the trio as human beings who executed their duties with ardor towards building India’s scientific communities. When did you decide to start a company, and where did your team get together? TD: We met as students in early 2016 at UC Berkeley. Mubhij had the idea of doing gene therapy for this particular form of colon cancer, and I was working a PhD thesis focused on overcoming physical barriers for gene delivery. In our study, published today in Science, we examined more than 124,000 reef-building corals and found that 89% of corals with trapped plastic had visual signs of disease – a marked increase from the 4% chance of a coral having disease without plastic. Globally, more than 275 million people live within 30km of coral reefs, relying on them for food, coastal protection, tourism income, and cultural value. With coral reefs already under pressure from climate change and mass bleaching events, our findings reveal another significant threat to the world’s corals and the ecosystems and livelihoods they support. In collaboration with numerous experts and underwater surveyors across Indonesia, Myanmar, Thailand and Australia, we collected data from 159 coral reefs between 2010 and 2014. In so doing, we collected one of the most extensive datasets of coral health in this region and plastic waste levels on coral reefs globally. There is a huge disparity between global estimates of plastic waste entering the oceans and the amount that washes up on beaches or is found floating on the surface. Our research provides one of the most comprehensive estimates of plastic waste on the seafloor, and its impact on one of the world’s most important ecosystems. Plastic litter in a fishing village in Myanmar. Kathryn Berry The number of plastic items entangled on the reefs varied immensely among the different regions we surveyed – with the lowest levels found in Australia and the highest in Indonesia. An estimated 80% of marine plastic debris originates from land. The variation of plastic we observed on reefs during our surveys corresponded to the estimated levels of plastic litter entering the ocean from the nearest coast. One-third of the reefs we surveyed had no derelict plastic waste, however others had up 26 pieces of plastic debris per 100 square metres. We estimate that there are roughly 11.1 billion plastic items on coral reefs across the Asia-Pacific. Just 4,000 steps a day can lead to better brain health

Walking more than 4,000 steps a day can improve attention and mental skills in adults 60 and older, according to UCLA research published December 12 in a preprint edition of the Journal of Alzheimer’s Disease. Various studies have found that physical activity is important in preventing cognitive decline and dementia in older adults. Cognitive decline occurs when people start having difficulty reasoning, processing and remembering. Brain volume and brain thickness — both measured by neuroimaging methods — are different ways of quantifying the health of the brain. Previous research shows physical activity correlates with higher volume in the hippocampus, a small, memory-critical region deep within the brain. “Few studies have looked at how physical activity affects the thickness of brain structures,” said Prabha Siddarth, the study’s first author and a biostatistician at the Semel Institute for Neuroscience and Human Behavior at UCLA. “Brain thickness, a more sensitive measure than volume, can track subtle changes in the brain earlier than volume and can independently predict cognition, so this is an important question.” During the two-year study, researchers recruited 29 people 60 and older with memory complaints. Researchers tracked their physical activity for seven days using accelerometers, and determined their average number of steps per day. Study participants were divided into one of two groups: those who walked more than 4,000 steps a day and those who walked fewer steps. The participants underwent a battery of neuropsychological tests and MRI scanning. People who walked more than 4,000 steps each day had a thicker hippocampus and thicker surrounding regions than those who walked less than 4,000 steps. Thickness in these regions correlates with better cognitive function. The more active group also had superior performance in attention and information-processing speed as well as executive functioning, a set of mental skills that allow people to make plans and achieve goals. Lower physical activity correlated with thinner brain structures and lower cognitive functioning. The researchers said future studies will track participants over a longer period in an effort to better understand the causes of thinning of the hippocampus. In addition to Siddarth, the paper’s other authors are Berna Rahi, Natacha Emerson, Alison Burggren, Karen Miller, Susan Bookheimer, Helen Lavretsky, Bruce Dobkin, Gary Small and David Merrill, all of UCLA. Radon gas is the leading cause of lung cancer in non-smokers. Children are most at risk of exposure in homes, especially during the winter months. (Shutterstock) Aaron Goodarzi, University of Calgary Back in 2013, in my first winter in my newly renovated Calgary home, I spent $45 on a radon testing kit. This was a logical move. I am an assistant professor in the Cumming School of Medicine at the University of Calgary, a cancer researcher and, above all, a radiation biologist specialising in the impact of radiation on DNA. I know that radon is the leading cause of lung cancer after cigarette smoking and that it causes up to 4,000 new lung cancer cases per year in Canada, more than 300 of them in Alberta. That same winter of 2013, I challenged several dozen of my colleagues — cancer researchers and physicians — to test their homes for radon gas. While my home seemed to be safe, many of my colleagues were not so lucky. They learned their homes contained radon levels far exceeding safe exposure. In the years that followed, my laboratory performed a study recruiting people as “citizen scientists” to test their homes for radon. We tested more than 2,300 houses in southern Alberta. The results, published earlier this year, were staggering: One in eight homes exceeded Health Canada’s acceptable radon level. Surprisingly, we also found that newer houses have the biggest radon problem. As a society, we have a well-recognised problem with environmentally-induced cancers. Testing your home for radon gas is an effective prevention strategy and radon is one of the most easily eradicated causes of lung cancer. The problem is most people do not know this. What is radon gas? Radon is a radioactive, invisible and odourless gas that comes from the natural breakdown of radium in the soil. It can seep into a building through foundation cracks and other openings. Mostly it seeps through the basements of homes, schools and workplaces. It can affect any Canadian building, both old and new, in nearly all inhabited geographical regions. The first major connection between radon gas and lung cancer was made in the 1970s, after Canadian uranium miners in Elliot Lake were found to have unusually high lung cancer rates. My daughter has Eczema which only shows up because of weather conditions (cold or hot, this is how it started), I just wanted to see what other moms use for lotions or something for their babies skin,  I was using the Aveeno with the navy blue lid but I noticed like red dots showing up after I put it on her (especially her back) so I want to stop using it but don't know which one to switch too.. Anyone have an suggestions?
