Lactobacillus fermentum 3872 genome sequencing reveals plasmid and chromosomal genes potentially involved in a probiotic activity | FEMS Microbiology Letters | Oxford Academic
OUP Academic
In this report we describe a Lactobacillus fermentum 3872 plasmid (pLF3872) not previously found in any other strain of this species. The analysis of the complete sequence of this plasmid revealed the presence of a gene encoding a large collagen-binding protein (CBP), as well as the genes responsible for plasmid maintenance and conjugation. Potential roles of CBP and a chromosomally encoded fibronectin-binding protein (FbpA) in probiotic activity are discussed.

How Stress Impacts Women
by Alanna Kuhn @ Blog - Kingston Naturopath, Kingston Hypothyroidism & Depression Treatments | Kingston Natural Route Health
Mon Jun 23 03:26:55 PDT 2014
Dr. Andrea Hilborn, NDOriginally published in the Kingston Whig-Standard
This article will focus on how stress intersects with being female. Our female physiology and female gender both impact the way women handle stress. Understanding how can help us manage it better. Men and woman all have three sex hormones: estrogen, progesterone and testosterone. These hormones affect all our body systems in myriads of ways. Imagine these three hormones as though they are radios playing different music. The volume of each radio can go up or down. Whichever radio is loudest is the one that has the strongest effect. During reproductive years, women's hormone levels are constantly fluctuating. In the first half of the cycle, estrogen is the dominant signal. In the second half, progesterone is the dominant signal. Estrogen has became a synonym for femaleness in everyday conversation, but progesterone is actually present at much higher levels. After menopause, sex hormones are in a steady state and no longer cyclically fluctuate. Healthy hormone levels are critical for longterm health. We know that having adequate progesterone levels throughout our reproductive years is protective against breast cancer, osteoporosis and heart disease, which are three of the big causes of mortality and morbidity. During the reproductive years, hormone imbalance can show up in many ways: irregular menstrual cycles, male pattern hair growth on face and body, changes in menstrual bleeding and PMS are some symptoms. In addition to the regular hormonal cycling that happens, women also undergo periods of major hormone shifts: puberty, pregnancy, and menopause. Menopause is the time from one year after a woman's last menstrual period continuing through the rest of her life. Perimenopause is the time up to one year after a woman's last menstrual period. The onset of perimenopause is when characteristic hormone-related signs show up. Common signs include heavier menstrual bleeding, shortening menstrual cycle, increased PMS symptoms and hot flashes. Perimenopause can begin as early as the mid-thirties. Stress impacts our hormones, and hormones impact our stress level. We know from animal studies that being in stressful situations over a long period of time decreases progesterone levels, and can inhibit ovulation. One theory of why this happens is that because the stress hormone cortisol is made out of the same material that progesterone is made out of, producing a lot of stress hormone reduces the ability to make progesterone. We also know that our sex hormone levels impact our ability to respond to stress. Cortisol levels increase during stressful situations. In the presence of higher progesterone levels, the increase in cortisol is smaller. Going through times of hormonal change can produce enormous stress. For example, if you start to experience intense hot flashes hourly, you will me worn out physically and emotionally, and less resilient to stressors in your everyday life. During perimenopause, estrogen levels start to spike and dip erratically, which can cause mood swings. Progesterone levels start to wane, causing greater sensitivity to stress. The combination of these two changes combine, reducing stress tolerance. I recommend keeping a menstrual cycle diary to see if your hormones and your stress level may be related. A blank diary can be found at www.natural-route.com/basal-body-temperature-chart Increasing your resilience to stress may mean treating hormonal imbalance. If you suffer from any of the symptoms discussed above, consider naturopathic care.
What's the Deal with Bio-Identical Hormone Therapy?
by Alanna Kuhn @ Blog - Kingston Naturopath, Kingston Hypothyroidism & Depression Treatments | Kingston Natural Route Health
Fri Jan 26 07:15:11 PST 2018
Bio-identical hormone therapy is NOW OFFERED at our clinic by Dr. Andrea Hilborn, ND. The hormones that may be prescribed are estrogen and progesterone in a cream or vaginal suppository.
Who is this for?
Bio-identical hormones are mainly used to treat the symptoms of menopause. The second most common use is as an anti-aging measure. They can also be used to treat just about any condition related to women's health: fertility, heavy bleeding, polycystic ovarian syndrome, acne, painful periods, and vaginal atrophy for example.
What are bio-identical hormones?
Bio-identical hormones, which are also called natural hormones, are exact replicas of the hormones our body produces. They may come as ready-made products or be custom compounded.
First, your body's hormone levels are tested, then we supplement the appropriate hormones until your symptoms are managed, without exceeding what your body would normally produce.
Taking hormones topically, rather than orally, allows you to take lower amounts of hormones and still receive the benefits. This is because the hormones entering through the skin do not have to pass through the liver before reaching the bloodstream.
The most important factor is to have a prescription that is based on the most up-to-date understanding of female physiology. Many people are under the impression that all menopause and perimenopause symptoms are caused by low estrogen, which is not the case.
Menopause
Menopause is the time from one year after the woman's final menstrual period to the end of her life. Levels of estrogen, progesterone and testosterone are lower, and at different levels in relation to one another. There is no more cyclic fluctuation of hormone levels. During the early years of menopause, women may still experience symptoms like hot flashes, which bio-identical hormones can help with. Bio-identical hormones are also used for anti-aging during this time of life.
Perimenopause
Perimenopause is the period of time just before menopause. It is characterized by wild fluctuations in hormone levels that can be compared to what women go through at adolescence. This stage begins in the mid-thirties to early-forties, and ends one year beyond final menstrual flow. This is the period of time when symptoms arise and can become life-disrupting.
The earliest symptoms of perimenopause are: new onset and/or longer menstrual flow; short menstrual cycle lengths of less than or equal to 25 days; new sore, swollen and/or lumpy breasts; new or increased menstrual cramps; new mid-sleep wakening; onset of night sweats especially around flow; new or markedly increased migraine headaches; new or increased premenstrual mood swings; and notable weight gain without changes in exercise or food intake.
Many women are inappropriately treated for fibroids when they experience heavy bleeding in perimenopause. Heavy bleeding is rarely caused by fibroids. The heavy bleeding is caused by high estrogen and lower progesterone. Hormonal treatment should be attempted before surgery.
During perimenopause, using bio-identical hormones can alleviate these symptoms.
Conventional Hormone Replacement Compared to Bio-Identical Hormone Replacement
Conventional hormone replacement therapy contains conjugated equine estrogens (CEE) (to mimic the action of estrogen) and progestins (to mimic the actions of progesterone).
Conjugated equine estrogens are derived from the urine of pregnant horses. There are a variety of molecules in the urine that stimulate the same cell receptors as the three different estrogens that we produce in our bodies. Premarin contains conjugated equine estrogens.
Widespread use of conventional HRT was discontinued when the Women's Health Initiative Study found that using HRT increased risk of cardiovascular events and cancer.
There are different forms of estrogen, which complicated the research findings. The estrogen-like molecules found in CEE modify estrogen receptors on breast cells in a cancer-promoting way. Synthetic progestins do too. Additionally, CEE contains at least one potent carcinogenic estrogen, 4-hydroxy-equilenin that promotes cancer by inducing DNA damage.
The progestins in conventional hormone replacement therapy include medroxyprogesterone acetate and norethrindone. Provera contains medroxyprogesterone acetate.
Prempro contains a combination of conjugated equine estrogens and medroxyprogesterone acetate.
Medroxyprogesterone acetate has breast cancer promoting properties, negates the heart protective actions of estrogen, causes blood vessels to constrict, and causes insulin resistance.
Using estrogen and bio-identical progesterone is a lesser risk than conventional HRT, but still imposes an increased risk compared to not using hormone replacement therapy.
Before booking an appointment to receive bio-identical hormone therapy, you should know:
We cannot prescribe bio-identical hormones to anyone with a history of breast cancer.
We use topical hormones and vaginal suppositories. We do not prescribe pills or injections.
We do not prescribe testosterone.
Every patient will have to have testing done, or bring results from within the last 6 months, and have repeat testing once per year.

Over the course of their lives, 1 in 3 women and 1 in 5 men will suffer an osteoporotic fracture; hip fractures related to osteoporosis result in death in nearly one third of cases.
Many people come to us with low bone mineral density despite having taken calcium supplements faithfully for decades. Calcium supplementation alone is not the answer. Our approach to low bone mineral density answers the question ‘how can we support the growth of healthy bones?’
We recommend nutritional supplements, provide you with a nutrition plan, and discuss physical activities. We also make sure that you aren’t being affected by any medical conditions that could decrease your bone mineral density, such as low stomach acid or hypothyroidism.
To make sure you are getting a good response to our treatments, we measure markers of bone turnover when you start treatment with us, and 6 months later.
To learn about your risk for fracture, check out this fracture risk assessment tool.

Gardening season is here again. I love gardening but this year, in a moment of overwhelm, I told my husband I did not want the hassle. It's a lot of work keeping up with a garden, and I was questioning the return on investment. Fortunately, he went ahead and did it anyways. He skipped the meticulous timing, the seed testing, the layout planning and just got some seeds and transplants in the ground. I'm glad because there are so many great things about gardening.

The scoop on probiotics
by Alanna Kuhn @ Blog - Kingston Naturopath, Kingston Hypothyroidism & Depression Treatments | Kingston Natural Route Health
Mon Jun 23 07:33:27 PDT 2014
by Dr. Andrea Hilborn, ND
Originally published in the Kingston Whig-Standard
Probiotic supplements are everywhere now – should you be taking one? Let me help you decode some of the information about these microorganisms that have be shown to have wide-ranging health benefits.
A probiotic is a bacteria that lives in or on us that produces a beneficial effect. Bacteria are found all over our skin, in our digestive system- just about everywhere. Bacteria have recently been discovered in urine (which was thought to be sterile), and even in the placenta. Many foods contain beneficial bacteria: yoghurt, cheese, sauerkraut, and tempeh for example.
A probiotic supplement is a commercial product that contains particular strains of probiotics in a capsule, powder or liquid. Probiotic supplements are getting more diversified all the time. For example, you can now purchase probiotic vaginal suppositories, which are used to relieve the symptoms of vaginal yeast infections.
Probiotic supplements are not safe for severely ill or immunocompromised people.
lThe Lactobacillus species are the most common probiotic supplements, and the best-studied. They reduce the risk of antibiotic-associated diarrhea by 36-44%. Lactobacillus rhamnosus appears to be most effective at reducing this risk. The Lactobacillus species can aide in digestion of lactose and glucose, and treat urinary tract infections.
Bifidobacterium is another common group of species found in probiotic supplements. They are usually packaged with Lactobacillus, rather than being on their own, and have many of the same effects as Lactobacillus. In combination, Lactobacillus and Bifidobacterium species can reduce the frequency of respiratory tract infections, by a surprising 42%.
Saccharomyces boulardii is actually a fungus, rather than a type of bacteria. Saccharomyces reduces the risk of Clostridium difficile by 61%.
Escheria coli NISSLE 1917 is a beneficial type of E.coli. It alters the immune response of intestinal cells, decreasing the effects of inflammatory bowel disease. It has also been genetically modified to deliver molecules that promote wound healing and inhibit the attack of HIV.
Studies are currently underway examining the possibility of creating a probiotic cocktail from a healthy person, to give to a sick person. The 'fecal microbiota transplant' is delivered by enema. Providing a complex mix of probiotics this way may help treat inflammatory bowel disease and recurrent C. difficile.
Our understanding of probiotics is in its infancy. We are starting to see that calling a type of bacteria good or bad may be too simplistic. Helicobacter pylori is associated with stomach ulcers. We thought for sure that it was a bad bacteria. Now we know that less than 20% of people infected with H. pylori will go on to have serious consequences.
Vegans and vegetarians should use extra care in choosing a probiotic. Some contain fish meal and many contain milk ingredients.
In my practice, I only recommend a probiotic if there is a specific symptom I am trying to treat, as opposed to advising people to take them even if they are completely healthy. In my opinion, if you are healthy, your bacteria are probably healthy. If you are ill, your bacteria probably suffer. Fixing your bacteria with a probiotic may help your health, and getting healthy probably creates a good environment for healthy bacteria to grow in. It's a chicken-and-egg relationship.

Top 5 Reasons Everyone Should Use a Meal Plan
by Alanna Kuhn @ Blog - Kingston Naturopath, Kingston Hypothyroidism & Depression Treatments | Kingston Natural Route Health
Fri Feb 09 04:21:55 PST 2018
By Anneke Aubert, Certified Culinary Nutrition Expert
These days everyone is “busy,” whether we like it or not, and so it becomes difficult to pay attention to our broader health, resulting in the majority of us only taking action and seeking help when something serious has already taken over our bodies. That is where the benefits of a meal plan come in, at least when it comes to your daily nutrition. If you just spend a little time at the beginning of each week prepping for the days ahead you wouldn’t end your week feeling so depleted, stressed or tired. How you ask? I have 5 detailed reasons below to justify why everyone should use a meal plan.
1. Get all your vitamins & nutrients from food!
Having a well thought out meal plan can help ensure you are getting a variety of nutrients and vitamins. That means you will feel more energized throughout the day, as well as reduce long-term effects of poor diet. Planning meals in advance means that you can add variety and nutritionally beneficial foods like fruits, vegetables, nuts, seeds, legumes to your daily diet. Knowing in advance what you are eating each day means you won’t be left feeling overwhelmed trying to decide what to eat for each meal. Without a doubt having an organized meal plan will lead to less or no consumption of processed, frozen or take out foods, which can also be costly to your wallet and health.
2. Save money on grocery bills!
Speaking of your wallet, meal planning is a great way to save money. Why you ask? Well, the fact that you are planning ahead means that you will only purchase what you need. This is especially important with perishable food items like fresh produce, dairy or meat that can spoil if you don’t have a plan for how you will use them. A meal plan means that at the end of the week you won’t be throwing away rotten spoiled food that you forgot you had in the back of your fridge. Another way meal planning can help reduce grocery costs is by selecting seasonal recipes or checking your local grocery store flyer to see what the weekly deals are and integrating these items into your plan. Cooking with the season usually means these foods are local and highly available, therefore making them more affordable than items that might be shipped from long distances. Your health can also benefit from eating locally and seasonally, as nutrient content is higher in foods that have not travelled great distances.
3. You may actually spend less time in the kitchen!
Proper meal planning by you or your nutrition expert may actually lead to you spending less time in the kitchen… if desired. I always ask my clients to choose one day a week where on that day they can reserve an hour or two to spend time prepping for the week ahead. An hour or two in the kitchen to some may sound like an eternity, but most will find that the tasks done in this time can feel like a form of therapy; wash, peel, cut, store, wash, peel, cut, store, repeat. Meal prep day for my clients is usually a time for them to not only make some recipes ahead of time, but a time to prep individual ingredients for snacks and meals that will be made later in the week. If you have family or friends that you think could reap the benefits of this therapeutic experience have them come help. The joy in cooking comes from sharing that food with the ones you care about.
4. Improve the efficiency of your day!
Having a well-organized meal plan can make your time in the kitchen a breeze. We all have those days where we find ourselves staring into the abyss of our fridge, trying to decide what we should make for dinner, and if we only had that one extra ingredient than we could make some awesome stir-fry, power bowl or casserole! Well, having a meal plan can be your saving grace when it comes to these tough decisions, as the meals are all planned out in advance taking the decision factor out of your morning, afternoon or evening. Not only will it help meal time prep be more efficient, a proper meal plan can help organize your fridge so you see clearly when you open the door to pull out your next snack or meal. Knowing what recipes are going to be made ahead of time and having ingredients prepped in advance, means you won’t be left wondering what your next meal is going to be, over and over again.
5. Advance recovery of health conditions or concerns!
A well researched personalized meal plan with recipes and foods that are beneficial to you and your family’s specific health conditions and concerns can significantly reduce symptoms of ongoing or potential health problems. Having a supportive health care provider, like a naturopath or your family doctor, who is an advocate for not only your current but future health concerns is so important when it comes to understanding what your individual body needs. Quite often I utilize doctor’s recommendations when creating a personalized meal plan for a client, making sure to research and pick specific foods that will be nutritionally beneficial to their health concerns and improve their body functions. Getting to know your body and what you are putting in it is the first step to taking responsibility for your own health!

Dr. Oz’s sterling reputation has helped him promote a number of weight loss supplements, but now his reputation is taking on some tarnish. In his own words, referring to the weight loss supplements he has promoted, “Oftentimes they don’t have the scientific muster to present as fact.” Over the next four columns, I’ll first talk about two weight loss supplement scams, then two weight loss supplement stars. Weight loss supplement scams don’t work and may cause harm. Weight loss supplement stars won’t cause you to start dropping pounds magically, but they may correct problems that can prevent weight loss.

Entrapment of Probiotics in Water Extractable Arabinoxylan Gels: Rheological and Microstructural Characterization
MDPI
Due to their porous structure, aqueous environment and dietary fiber nature arabinoxylan (AX) gels could have potential applications for colon-specific therapeutic molecule delivery. In addition, prebiotic and health related effects of AX have been previously demonstrated. It has been also reported that cross-linked AX can be degraded by bacteria from the intestinal microbiota. However, AX gels have not been abundantly studied as carrier systems and there is no information available concerning their capability to entrap cells. In this regard, probiotic bacteria such as Bifidobacterium longum have been the focus of intense research activity lately. The objective of this research was to investigate the entrapment of probiotic B. longum in AX gels. AX solution at 2% (w/v) containing B. longum (1 × 107 CFU/cm) formed gels induced by laccase as cross-linking agent. The entrapment of B. longum decreased gel elasticity from 31 to 23 Pa, probably by affecting the physical interactions taking place between WEAX chains. Images of AX gels containing B. longum viewed under a scanning electron microscope show the gel network with the bacterial cells entrapped inside. The microstructure of these gels resembles that of an imperfect honeycomb. The results suggest that AX gels can be potential candidates for the entrapment of probiotics.
